Abstract Modafinil is a psychostimulant drug used widely for the treatment of narcolepsy, which also has additional positive effects on cognition. Here, we investigate the effects of modafinil on behavioral performance and synaptic plasticity in rats. Improved acquisition of water maze task was observed for animals that underwent chronic treatment with modafinil. We found that the distance travelled and escape latency were reduced after the first day in chronically-treated rats, compared to controls. Importantly, the swim velocity was similar for both groups, excluding pharmacological effects on motor skills. We also found that modafinil increases synaptic plasticity in the dentate gyrus of urethane-anaesthetized rats; modafinil induced a robust augmentation of the population spike, evident after application of 2 bursts of 200Hz, high-frequency stimulation. Furthermore, the modafinil-dependent enhancement of postsynaptic potentials correlated selectively with theta rhythm augmentation. We propose that modafinil may facilitate spatial orientation via increased theta-related hippocampal plasticity.

Running enhances neurogenesis, learning, and long-term potentiation in mice.

Running increases neurogenesis in the dentate gyrus of the hippocampus, a brain structure that is important for memory function. Consequently, spatial learning and long-term potentiation (LTP) were tested in groups of mice housed either with a running wheel (runners) or under standard conditions (controls). Mice were injected with bromodeoxyuridine to label dividing cells and trained in the Mor...

Effects of parental morphine addiction on long term potentiation of the perforant path to dentate gyrus in rat offsprings

Background: Evidences show that parental morphine addiction impairs CNS development, learning and memory in offsprings. Since long term potentiation (LTP) is a cellular mechanism of learning and memory, in this study the effect of parental morphine addiction on LTP induction in dentate gyrus by high frequency stimulation of perforant path was assessed. Materials and methods: In this experiment...

Effect of Clenbuterol Administration into the Basolateral Amygdala on Synaptic Plasticity in Dentate Gyrus Granule Cells in Male Rats

Background and purpose: Neural circuits and neurotransmitter systems within the basolateral amygdala (BLA) play roles in forming emotional memory through communication with the hippocampus. Therefore, in this study, the role of these neural circuits on synaptic plasticity was investigated by agonist injection of beta-adrenergic receptors into the BLA. Materials and methods: In this experiment...

Emotional and cognitive information processing: relations to behavioral performance and hippocampal long-term potentiation in vivo during a spatial water maze training in rats.

Emotionality as well as cognitive abilities contribute to the acquisition and retrieval of memories as well as to the consolidation of long-term potentiation (LTP), a cellular model of memory formation. However, little is known about the timescale and relative contribution of these processes. Therefore, we tested the effects of weak water maze training, containing both emotional and cognitive d...

Cumulative long-term potentiation in the rat dentate gyrus correlates with, but does not modify, performance in the water maze.

The electrically induced increase in hippocampal synaptic strength known as long-term potentiation (LTP) is thought to involve some of the same mechanisms as those mediating information storage during spatial learning. Physiological saturation of synaptic weights might therefore be expected to occlude spatial learning. In support of this, Castro et al. (Castro CA, Silbert LH, McNaughton BL, Bar...
